Cholinesterase based amperometric biosensors for assay of anticholinergic compounds
نویسنده
چکیده
Biosensors are analytical devices being approachable for multiple analytes assay. Here, biosensors with intercepted acetylcholinesterase (AChE) or butyrylcholinesterase (BChE) are presented as tool for assay of anticholinergic compounds such as pesticides, nerve agents and some natural toxins. Principle of assay is based on evaluation of cholinesterase activity and its pertinent decrease in presence of analyte. Nerve agents, pesticides, anticholinergic drugs useable for treatment of Alzheimer's disease as well as myasthenia gravis and aflatoxins are enlisted as compounds simply analyzable by cholinesterase biosensors.

Determination of serum cholinesterase activity and dibucaine numbers by an amperometric choline sensor.
Serum cholinesterase activity and the dibucaine numbers have been determined by using a hydrogen peroxide electrode and the enzyme choline oxidase immobilized on a nylon net. The analysis procedure is extremely simple and very fast allowing 30 cholinesterase determinations per hour. Both cholinesterase activity and dibucaine number measurements could be performed in 5 min and by using serum sam...

Since the appearance of the first amperometric biosensors based on the coupling of oxidase enzymes and relying on the final detection of H2O2, it was clear that one of the major problems related to this kind of configuration was due to the high overpotential needed for H2O2 oxidation (ca. 0.7V vs. Ag/AgCl). At this potential in fact, many electroactive substances (i.e. ascorbic acid, uric acid ...
متن کاملCritical Evaluation of Acetylthiocholine Iodide and Acetylthiocholine Chloride as Substrates for Amperometric Biosensors Based on Acetylcholinesterase
Numerous amperometric biosensors have been developed for the fast analysis of neurotoxic insecticides based on inhibition of cholinesterase (AChE). The analytical signal is quantified by the oxidation of the thiocholine that is produced enzymatically by the hydrolysis of the acetylthiocholine pseudosubstrate. The pseudosubstrate is a cation and it is associated with chloride or iodide as corres...
متن کاملSynthesis, X-Rays Analysis, Docking Study and Cholinesterase Inhibition Activity of 2,3-dihydroquinazolin-4(1H)-one Derivatives
In search of potent cholinesterase inhibitors, we have carried out the synthesis and biologically evaluation of various benzaldehyde based 2,3-dihydroquinazolin-4(1H)-one derivatives. In vitro assay results revealed that all the synthesized compounds showed activity against both enzymes (AChE and BChE) and in few cases, the inhibition activity was even higher than or comp...
متن کاملChemometric exploration of an amperometric biosensor array for fast determination of wastewater quality.
Four wastewater samples of different treatment qualities; untreated, alarm, alert and normal, from a Swedish chemi-thermo-mechanical pulp mill and pure water were investigated using an amperometric bio-electronic tongue in a batch cell. The aim was to explore enzymatically modified screen-printed amperometric sensors for the discrimination of wastewater quality and to counteract the inherent dr...
